MOTS-c, SS-31, NAD+ Peptide Protocols

All right, so everybody's talking about MOTS for fat loss and energy, but almost nobody is using it correctly. You see, using MOTS-calone is like putting racing fuel in a car with a cracked engine block. It doesn't matter how good the fuel is if the engine can't use it.

And here's the pattern that I keep seeing. People spend hundreds of dollars on MOTS-cexpecting dramatic fat loss, increased energy, and better recovery. They take it for 8 to 12 weeks, and what do they get? Maybe they drop a few pounds. Maybe their energy improves slightly. But nothing close to what the research suggests is possible.

And the reason has everything to do with mitochondrial health and how these peptides actually work together. So in this post, I'm going to show you the correct protocol based on where you're starting from. But before I can do that, I need you to understand how these peptides actually work, because that's the only way you'll understand why the common approach is backwards and what to do instead.

Understanding Mitochondria

All right, so let's start with your mitochondria. Your body makes ATP primarily in the mitochondria through the electron transport chain. Dragon Pharma NAD+ is critical for this process. Now, SS31 repairs damaged mitochondrial membranes. It binds to the cardiolipin, stabilizes the inner membrane, and reduces the leakage of electrons during that energy production process by 40 to 60%. So why does this matter? Well, because damaged mitochondria leak electrons. They create oxidative stress and can't produce ATP efficiently. So think of this as a mechanic that fixes structural damage: cracks, leaks, broken components. That's what SS31 is doing.

And MOTS-c is much different. It works by activating AMPK, your metabolic master switch, and improves your insulin sensitivity and reduces ROS production. And here's the key insight. MOTS-c actually reduces the thing that damages mitochondria in the first place. So this is both a performance upgrade and a protective system that prevents future damage.

HOW TO USE MOTS AND SS-31 PEPTIDES TOGETHER
Two Approaches
1. Use MOTS first, then SS31 only if needed.

The logic here is that MOTS reduces ROS production, which damages mitochondria in the first place. By improving metabolic efficiency, you prevent the damage that makes SS31 necessary. This makes perfect sense if you're younger, relatively healthy, and wanting preventative optimization. In this approach, MOTS-c becomes what some call a forever peptide. You take it indefinitely as a foundational upgrade. SS31 is only used if needed after four to eight weeks on MOTS-c. Basically, you take MOTS, you don't really see the benefits, so you use SS-31 to repair the damage.

2. Repair first with SS-31, then optimize with MOTS-c.

The logic here is if you already have significant accumulated mitochondrial damage, repair it first so MOTS signals can work properly. Because damaged mitochondria can't respond effectively to MOTS optimization signals. Fix the broken engine before installing the performance upgrades. This makes sense for people who are older with declining energy, poor recovery, metabolic dysfunction. Because by your 40s, you likely have 10 to 20 years of accumulated mitochondrial damage.

So why does this contradiction exist? Well, it comes down to prevention versus repair. One approach prevents future damage. The other addresses existing damage.

So here's my position. Men and women between 35 to 55 experiencing energy decline, slower recovery, and metabolic issues, I believe the repair first approach makes more sense. By the time you're in your 40s with these symptoms, you likely have accumulated damage that needs addressing. But if you're younger, healthier, and want preventative optimization, the MOTC first approach makes perfect sense.

PROTOCOLS OF NAD+ SS-31 and MOTS
1. Repair-First Protocol

Use this if you already have metabolic dysfunction.
Weeks 1–4: Repair Phase
    SS-31: 1–2 mg daily
      NAD+: 100 mg, 3× per week

      During these first four weeks, you’re repairing accumulated mitochondrial damage and giving your cells extra fuel with NAD+.
      Weeks 5–12: Optimization Phase
        Stop SS-31 and switch to MOTS-C: 10 mg per week, split into 3 injections
          Continue NAD+: 100 mg, 3× per week

          Here you’re optimizing the mitochondria you repaired in the first phase.

          After Week 12
            Take 4–8 weeks off completely.
              You can repeat this cycle 2–3 times per year.

              2. Prevention-First Protocol

              Use this if you’re younger or generally healthy.
              Foundational Phase (Ongoing)
                MOTS-C: 10 mg per week, split into injections on Monday, Wednesday, Friday
                  NAD+: 100 mg, 3× per week

                  This helps prevent damage and keeps metabolism optimized.
                  Note: You must cycle off NAD+ after about 12 weeks to avoid developing resistance.

                  Assessment at Weeks 4–8

                  Ask yourself:
                  Do you still have significant fatigue, poor recovery, or metabolic issues?
                    If NO:
                    Continue with MOTS-C + NAD+ as maintenance.
                      If YES:
                      Add SS-31 at 1–2 mg daily for 4–8 weeks, while continuing:
                        MOTS-C: 10 mg/week
                          NAD+: 100 mg, 3× per week

                          After finishing the SS-31 peptide cycle, return to the regular MOTS-C + NAD+ maintenance plan.

                          5-AMINO-1MQ PEPTIDE

                          Now there is one more thing that we can add here to kind of put icing on the cake for both of these protocols. And that's a peptide called 5-Amino-1MQ. Really quick, just so I can kind of visually explain this entire system. If I was to look at the energy production as an engine in a car, we're going to look at SS31 as the mechanic that's going to repair any problems with it. Then we're going to look at MOTS-c as like throwing a supercharger on it, right? NAD plus is the fuel. And then if we wanted to kind of put icing on the cake, you would look at 5-Amino-1MQ as a way to prevent waste of energy, to recycle that energy and create more efficiency in the system.

                          So I guess the best way to explain the 5-Amino-1MQ if you were a mechanic would be like if you had an exhaust leak, 5-Amino-1MQ would actually repair that. For context, if we want to further optimize, we would add in the 5-Amino-1MQ as a way to prevent energy waste and make sure that the NAD plus that we use can more easily be recycled again for further use.

                          WHAT WE SHOULD EXPECT FROM CYCLE

                          Okay, so let's move into what you should expect. For the repair first protocol, weeks one through four, your baseline energy starts improving and recovery gets much faster. Then by weeks eight to 12, you should feel a significant difference in energy, performance, and body composition.

                          For the prevention first protocol, weeks one through four, you'll see a gradual metabolic optimization and energy stabilization. Then week eight and beyond, you'll notice differences in sustained energy, improved body composition, and better recovery. But here's the key. If you don't feel substantial changes, either your dosing is wrong or your foundational habits need work. We can spend all this time, money, and effort working with peptides to try to repair your energy production system. But if you're not giving your body what it needs through sleep, food, and exercise, this whole effort is kind of a moot point. These protocols enhance a solid foundation. They do not replace it.

                          Now, before you run off and start this, you need to understand cycling. For the repair first protocol, do 12 weeks total, then take four to eight weeks off. And like I said earlier, this matters because NAD plus supplementation can suppress your body's natural NAMPT production. That's the enzyme that makes NAD plus. Long-term supplementation could actually make your body worse at producing NAD plus naturally, and that's a big problem.

                          For the prevention first protocol, MOTS-c can be taken long-term with periodic breaks, four weeks off every12 to 16 weeks. NAD plus still needs cycling, eight to 12 weeks on, four to eight weeks off. And then for SS31, you're just going to use it for four weeks. weeks, then come off and kind of use it as needed for repair.

                          CONCLUSION

                          So which protocol is right for you? Well, to give you kind of a rule of thumb:

                          If you're 35 to 55 with declining energy and metabolic issues, the repair first protocol is likely what makes better sense for you.

                          If you're younger, healthier, you don't have energy production issues or fatigue, and you want prevention, definitely go the MOTS-c route, and that'll be the logical choice for you.
